Description Sequencing of the new generation has enabled in recent years to analyze in detail the proportion of mutations in malignant diseases. Analysis of mutations in patients with acute myeloid leukemia then enabled to describe clonal composition and development in this disease. Studies using xenotransplantation of patients' primary cells into immunodeficient mice have also contributed significantly to the study, making it possible to monitor clonal changes in artificially created conditions. Clonal selection in mice is controlled by the inherent proliferative capacity of individual (sub) clones, but the use of mice expressing human cytokines or implanted humanized tissue also demonstrates the profound influence of the microenvironment. This has been shown to be critical for attachment of acute myeloid leukemia with a favorable prognosis, eg, RUNX1-RUNX1T1, CBFB-MYH11, NPM mutation without FLT3-ITD, bielelic mutation CEBPA. The aim of this review is to present up-to-date possibilities of simulating acute myeloid leukemia in vivo and to summarize the most important knowledge of clonal composition and pathogenesis of this disease obtained from xenotransplantation studies.
