Description Background. Monoclonal gammopathies (MG) are characterised by presence of different numbers of clonal plasma cells (PC), lower in monoclonal gammopathy of undetermined significance (MGUS) and higher in multiple myeloma (MM). Discrimination between normal polyclonal PC (N-PC) and abnormal clonal PC (A-PC) is important in time of diagnosis and especially after treatment (transplantation etc.). Expression of surface markers CD19 and CD56 on PCs is widely used for this discrimination, but unfortunately in some cases is not enough sufficient. Aims. Verification and validation of 8-color flow cytometry protocol for clonality assessment of PC and B cell subsets in MGs.Analysis using combination of surface and intracellular markers in 8-color setting can improve detection even small clone of pathological PCs and together with B cell assessment can be very important for determination of diagnosis and monitoring after treatment.
