This study investigates the interaction between delta opioid receptors (DOR) and inflammatory mediators in primary sensory neurons. The project focuses on how cytokine IL-6 and chemokine CXCL12 influence DOR priming, receptor trafficking, and localization at the plasma membrane under inflammatory conditions. Two complementary approaches—density gradient fractionation and antibody-based immunoisolation—will be used to assess receptor distribution between membrane and endosomal compartments. The study aims to clarify molecular mechanisms regulating DOR availability at the neuronal surface and to establish reliable methods for detecting receptor trafficking. The results will provide a methodological and conceptual basis for future research into the modulation of peripheral opioid analgesia by inflammatory mediators.
