Description The transverse (t-) tubular system is formed by invaginations of the surface membrane that brings excitation deep inside the cell. In our previous study [1] we formulated a mathematical model of human ventricular cell that included the t-tubular system. In the present study we improved the model by introducing description of new details based on available experimental data related to detailed morphology and properties of ionic currents of human ventricular myocytes. Further, the t-tubule compartment was partitioned into concentric cylindrical segments to describe the ion diffusion within t-tubular lumen. The simulations showed that if the fraction of tubular Ca2+ pump is low (lower than 20%) and the fraction of tubular Ca2+ channels (L-type) is high (higher than 60%) the activity induced transient reductions of Ca2+ concentration in human t-tubules might significantly lower Ca2+ entry into the cell and thus reduce the intracellular Ca2+ transients that control the strength of cellular contraction.
