Folding of approximately 2 m of human DNA molecule into a cell nucleus requires well-organized and tightly regulated cooperation of several protein complexes. One of the most important factors organizing DNA into higher-ordered structures such as chromatin loops are SMC (Structural Maintenance of Chromosomes) complexes, involving Cohesin, Condensin and SMC5/6. This project is dedicated to the less famous but not less important SMC5/6 complex, which plays important roles in the DNA repair and replication. Using highly sophisticated and complex chromatin conformation capture (CCC) methods, we would like to study the formation of chromatin loops that are created and hold by the SMC5/6. Based on our results, we would like to create a 3D model to visualize the organization of the chromatin in the cell nucleus, which would help to understand the dynamics of chromatin organization and its importance for the fundamental processes in a cell, such as DNA replication and repair.
