Description Tumors of urinary bladder, kidneys and prostate are very often in the Czech Republic and are responsible for 3% of cancer related death. This is because there is no reliable and sensitive biomarker. MicroRNAs (miRNAs) are short non-coding RNAs that post-transcriptionally regulate gene expression of more then one-half of human genes including important oncogens and tumor suppressors. Deregulated expression profiles of miRNAs have been described in tumor tissues as well as in body fluids. Recently, several papers were published analyzing miRNAs in urine in order to diagnose early stages of urological tumors. Before it will be possible to use urine miRNAs as new biomarkers, it is necessary to solve several problems and methodical approaches. The main aim of our study was to analyze expression of miR-125b and miR-21 in supernatant of urine of healthy donors using qRT-PCR and set the main parameters of analysis, find an appropriate endogenous control and vereficate the stability of miRNAs. Subsequently, the methods of detection were optimized.
