Capillary electrophoresis with contactless conductivity detection as a promising alternative for kinetic and inhibition studies of beta-secretase

Description Alzheimer’s disease is a major form of dementia which prevalence increasing with aging population. Despite intensive research and ongoing clinical trials, effective causal or symptomatic treatment of Alzheimer’s disease is not yet available. The disease is manifested with decline in memory and cognitive functions leading to progressive disorientation in time and place, and eventually death. The brains of the patients are histopathologically characterized by cerebral plagues laden with ß-amyloid peptide. This peptide originates from proteolytic cleavage of amyloid precursor protein by ß-secretase. Consequently, inhibition of ß-secretase might be effective treatment of Alzheimer’s disease slowing down the disease progression. Accuracy of inhibition assays might be compromised by use of labeled enzyme substrates or fluorescence quenching in the reaction mixture thus separation-based assay by means of capillary electrophoresis coupled with contactless conductivity detection was developed.
