Description We studied programmed cell death (PCD) in two promonocytic cell lines - human U937 cells and chicken v-myb-transformed monoblasts. PCD was induced using three cytotoxic agents: camptothecin, cycloheximid and arsenic trioxide. Cell death was analyzed by measurement of mitochondrial membrane potential, DNA fragmentation, F-actin depolymerization and caspase activity. We found that camptothecin-treated BM2 cells undergo caspase-independent cell death while similarly treated U937 cells undergo caspase-dependent PCD. This suggests that v-Myb that is constitutively expressed in BM2 cells may prevent caspases from activation thus inducing alternative caspase-independent cell death pathways.
