Description The aim of this study was to type L. monocytogenes isolated from dairy products and on the base of L. monocytogenes characteristics confirm the occurrence of persistant strains in the environment of dairy plants. A total of 102 isolates of L. monocytogenes obtained from dairy product samples originating from producers or retail outlets in the Czech Republic were tested. L. monocytogenes isolates were characterized by serotyping using slide agglutination, the results were confirmed by multiplex PCR assays, and by restriction analysis using pulsed-field gel electrophoresis (PFGE). Antimicrobial resistance was tested by the disc diffusion method. Strains belonged to serotypes 1/2a (84 % of isolates) and 1/2b (16 % of isolates). All of them were sensitive to the antibiotics tested, except oxacillin, cefotaxime and sulphonamides. Using macro-restriction analysis, 13 different pulsotypes were detected. Strains of the same origin always belonged to one or a few specific clones These results demonstrate the considerable heterogeinity of L. monocytogenes within one serotype. The presence of some clones L. monocytogenes specific for individual products examinated during several years indicates the possible occurrence of persistant strains in the dairy plants.
