The objective of this proposal is to ident´fy and characterize autologous T cells that are able to specifically destroy tumor cells from patients with metastatic melanoma. Melanoma cell apoptotic bodies-pulsed dentritic cells will be used to activate autologous T cells. Melanoma-specific T cells will be activated andsorted out by flow cytometer. Clonality of these activated T cells will be studied using cDNA library and direct sequenciong of T cell receptor beta genes. Melanoma-specific T cell clones will be identified, quantified, expanded, tested for cytotoxicity against original melanoma cells, and potentially use in future clicical studies.
