Klíčová slova yeast protoplasts; actin cytoskeleton
Popis Restoration of actin cytoskeleton was studied using cells converted into protoplasts. In fresh protoplasts both cytoplasmic and nuclear microtubules desintegrated. Similarly actin cables lost their integrity while patches became evenly distributed under the protoplast surface. A collapse of the cytoskeleton was caused by hypertonic stress when the cells were transferred to osmotic stabilizers. Microtubules and actin structures were renewed differently: cytoplasmic microtubules followed by nuclear microtubules appeared after 30-60 min of incubation. In S. cerevisiae and S. japonicus protoplasts actin appeared only in the form of patches:actin cables were seen neither in spherically nor polarly growing protoplasts. The cables appeared at the stage when the cell wall had been completely regenerated and a bud became apperent.
